Sažetak
Pre-embedding double immunocytochemical methods were applied at the electron microscope level to study the distribution of the receptor for substance P, the neurokinin 1 (NK1) receptor, on myenteric neurons in the guinea-pig small intestine and the correlation between the receptor distribution and presence of substance P immunoreactive axons. Tissue obtained from guinea pigs was incubated in culture medium for 1 hour and then fixed in 4% paraformaldehyde and 0.05% glutaraldehyde in phosphate buffer. Wholemount preparations of longitudinal muscle and myenteric plexus were labelled with antibodies against NK1 receptor and substance P. One of the antigens was localised with the DAB peroxidase reaction product, while the other antigen was visualised using silver intensified 1 nm immunogold technique. Three myenteric neurons expressing strong immuno-gold immunoreactivity were sectioned and every tenth to fifteenth section was analysed. Low power electron micrographs (x 8000) of the entire cell body were taken and appositions between the cell and a) substance P- immunoreactive terminals, b) non-immunoreactive nerve terminals and c) glial cells (or their processes) were examined. The length of the apposition was measured (using the image analysis program) and the number of silver particles which came into contact with the post-synaptic membrane at the apposition were counted. The NK1 receptor was evenly dispersed over the entire somatic membrane. The abundance of the receptor distribution was similar at substance P immunoreactive terminals, non- substance P reactive terminals and glial appositions. Also, there was significantly less NK1 receptor- immunoreactivity at the synaptic specialisation zone than at the apposition regions peripheral to it.
