Sažetak
Background. Paraoxonase (PON) is a HDL-associated esterase with antioxidative and antiatherogenic properties. Polymorphisms in promoter and coding regions of the pon genes affect PON activity. The aim of this study was to determinate the genotype distribution and frequencies of the alleles C and T for -108C>T polymorphism of pon1 gene and the alleles S and C for S311C polymorphism of pon2 gene in healthy Croatian population, and to determinate an ideal amplification temperature for each polymorphism. Methods. The study included 87 healthy unrelated individuals. Genomic DNA was isolated from EDTA- anticoagulated blood by FlexiGene DNA kit. Ideal amplification temperature was experimentally determined. Polymorphisms were determined by the polymerase chain reaction followed by restriction fragment length polymorphism analysis (PCR-RFLP). Results. The following genotype frequencies were determined: 21% CC, 57% CT and 22% TT for -108C>T polymorphism of pon1 gene, and 52% SS, 44% SC and 4% CC for S311C polymorphism of pon2 gene. The most frequent alleles were: allele T (51%) for -108C>T polymorphism of pon1 gene and allele S (74%) for S311C polymorphism of pon2 gene. Ideal amplification temperature for -108C>T polymorphism was 69°C and for S311C polymorphism 55°C. SigmaStat for Windows version 3.0 was used for statistical analysis. Allele and genotype frequencies were compared using the Chi-square test. Conclusions. The most frequent genotype in healthy Croatian population for -108C>T polymorphism of pon1 gene was CT, and for S311C polymorphism of pon2 gene was SS.
