Sažetak
Nitrofurans, as 5-nitrofuraldehyde derivatives, are an important group of synthetic antibiotics with broad spectrum activity. Due to their antibacterial and pharmacokinetic properties, they have been widely and effectively used for the prevention and treatment of various infections caused by certain pathogenic bacteria, and as an addition to animal feed as a growth promoter. However, nitrofurans have proven to be potentially carcinogenic and mutagenic. Thus, their use in veterinary medicine has been prohibited in the Republic of Croatia pursuant to Council Regulation 37/2010/EU of 22 December 2009 on pharmacologically active substances and their classification regarding maximum residue limits in foodstuffs of animal origin—Table 2–List of prohibited substances for which the MRL cannot be established. Regardless of the ban, the EU requires strict control of nitrofurans and their metabolites due to their high toxicity and the possibility of illegal animal treatment. In accordance with Commission Decision 2003/181/EC, the EU has set a Minimum Required Performance Limit (MRPL) for nitrofurans and their metabolites of 1 μg/kg, for every analytical technique used for the determination of nitrofurans and their metabolites in animal tissues and foodstuffs of animal origin. The LC- MS/MS method for the simultaneous detection of four nitrofurans (furazolidone, furaltadone, nitrofurantoin, nitrofurazone, respectively: AOZ, AMOZ, AHD and SEM, and their metabolites) was validated according to the validation method criteria of Commission Decision 2002/657/EC. Due to the short half-life of the parent compounds of nitrofurans, formed tissue-bound metabolites were determined as marker residues for nitrofurans. The following validation parameters were set: specificity, linearity, accuracy (expressed as recovery) 97.6–101.9%, limit of decision (CCα) 0.28–0.34 μg/kg, detection of capability (CCβ) 0.34–0.44 μg/kg, precision with CV 9.1–11.7%. A total of 60 muscle samples (20 beef, 15 pork, 15 poultry, 10 fish) were analyzed for residues of nitrofuran metabolites. The range of concentrations of quantified results was (μg/kg): AOZ 2.8x10-4–0.37, AMOZ 5.7x10-4–0.26, AHD 8.3x10-3–0.29, SEM 7.3x10-4–0.28. The presented results were lower than the MRPL for nitrofurans of 1 μg/kg. Accordingly, the results suggested that the tested meat samples are free of nitrofuran residues and therefore pose no concern for human health.
