Sažetak
Honey is a sweet, thick juice produced by the honeybees from the nectar which they collect from fresh flowers or by collecting sweet secretion (honeydew) of some insects. Honey contains amylolytic enzyme diastase. Diastasis activity is directly connected with the age, quality, storage and heat treatment of honey. The diastase enzymes are α - , β - and γ – amylase which hydrolyze the starch. Starch and three-iodide make a complex, in which three- iodine ion forms redox buffer and iodide ions. Due to the hydrolysis of starch with diastasis enzymes from honey, the concentration of ions in three-iodine is changed in the ratio of concentration redox couple three - iodide / iodide. This change causes a change in the redox potential that can be measured with platinum redox sensor, since this change in ratio of redox couple three – iodide / iodide is described by the Nernst expression. Potential increase, as a result of increasing the concentration of ions in three-iodide ion in ratio of concentrations in redox couple three-iodide/ iodide is in direct correlation with the concentration of diastase in honey. In this paper, we used a platinum redox sensor and direct potentiometric method. Additionally, the diastase activity was measured by Schade method (standard procedure) and the results were compared. Results showed good correlation.
