Sažetak
Surface display of proteins in bacteria and yeasts has proved to be an interesting method of protein immobilization for different purposes. Generally, proteins of interest are genetically engineered by fusing them to autochthonous cell wall proteins, or parts of autochthonous proteins directing them to the wall and anchoring them at the cell surface. In this way chemical reactions used to immobilize proteins to solid matrices which are often laborious and affect protein conformation and activity, are by-passed. Yeasts incorporate unusually high number of different autochthonous proteins into the cell wall. Some of these proteins have roles in building, maintaining, and remodeling cell wall material while the others are involved in the communication of the cell with the surrounding molecules or other cells. Yeast cell wall proteins can be noncovalently (Scw-proteins) or covalently (Ccw-proteins) bound to β-1, 3-glucan, either through GPI-anchors and β-1, 6-glucan, or by alkali labile ester linkages between γ-carboxyl groups of glutamic acid and hydroxyl groups of glucoses (Pir-proteins). Mechanisms of cell wall incorporation which can efficiently be exploited for surface engineering of yeast cells will be discussed.
