Sažetak
Planktonic diatom species of the genus Pseudo-nitzschia are common in oceanic and neritic plankton worldwide. Fourteen Pseudo-nitzschia species have been observed to produce domoic acid, a neurotoxic amino acid responsible for Amnesic Shellfish Poisoning (ASP). Therefore considerable scientific interest in its ecology and taxonomy has been generated. The taxonomic composition of Pseudo-nitzschia species in the Adriatic Sea is still unknown. For detailed taxonomical identification within the genus Pseudo-nitzschia in the eastern Adriatic sea diatom frustules were acid cleaned and analysed on a Zeiss Axiovert inverted microscope and on a transmission electron microscope (FEI Morgagni 268D). The examination of ultrastructural features revealed the presence of six species, and their distribution in phytoplankton assemblages along the Adriatic coast was determined. The species Pseudo-nitschia caciantha was for the first time recorded for the Adriatic sea. P. calliantha, already described as dominant in phytoplankton assemblages in the Northern Adriatic, was found also on four different stations in the southern and middle Adriatic. P. pseudodelicatissima distribution indicated this species as a permanent constituent of phytoplankton assemblages in the coastal part of the Adriatic Sea. P. mannii was found in the Northern and in the middle Adriatic. From the seriata group, P. pungens was found only in Northern Adriatic, while P. fraudulenta was found with low contribution to phytoplankton assemblages also in the middle Adriatic, and P. subfraudulenta only in the southern stations. Monoclonal cultures of Pseudo-nitzschia cells (isolated from May and October samples from NP Telašćica) were established and molecular analysis on different molecular markers (18S, ITS1-5, 8S-ITS2, 28S) has been performed. Phylogenetic analysis resulted in the identification of P. mannii and the potentially toxic P. fraudulenta. Statistical phylogenetic networks demonstrate genetically unique populations for both species in the middle Adriatic. Studies elucidating the nutrient and physical requirements, the biotic interactions of Pseudo-nitzschia species as well as species composition on regional and global scales are essential for the prediction and prevention of its mass occurrence and resulting toxicity.
