Sažetak
Type 1 diabetes (T1D) is an autoimmune disease that results from T cell-mediated destruction of insulin-producing β-cells. Disease transfer has been obtained with both CD4+ and CD8+ T cell clones after injection into immuno-incompetent recipient mice, which speaks in favor of T cells being the main therapeutic target in T1D. We have postulated that targeting activated T cells by using a combination of antibodies specific for CD25, CD154 and CD8 could prevent diabetes onset and restore normoglycemia in newly hyperglycemic NOD mice. We have shown that an antibody combination therapy consisting of CD8-, CD25- and CD154-specific antibody can reverse hyperglycaemia in NOD mice. Unfortunately, our data from Elispot analysis show that there is still much to learn when it comes to suitable methods to assess this research area. This preliminary study opens possibilities for further investigation on the synergistic possibilities of using several antibody based agents
