Introduction: Reverse transcription-polymerase chain reaction (RT-PCR) based assays are being used to study tumor dissemination and minimal residual disease. Tyrosinase mRNA, which is expressed only in melanocites and melanoma cells, has shown high specificity as a marker for RT-PCR detection of circulating melanoma cells. Aim: To evaluate RT-PCR for tyrosinase mRNA assay for the detection of circulating melanoma cells in melanoma patients. Methods: Blood samples were taken from 70 patients with melanoma in all clinical stages of the disease. RNA was isolated from mononuclear cell fraction of the blood and reversely transcribed into the complementary DNA (cDNA). The cDNA was assayed by PCR for the expression of tyrosinase. Peripheral blood smaples from 30 healthy volunteers were used as Controls. Results: The threshold for detection of tyrosinase was set low enough to detect 50 melanoma cells in 10 mL of healthy volunteer blood. Positive results for tyrosinase were obtained in 1 (5%) out of 19 stage I patients, 2 (10%) out of 21 stage II patients, 2 (13%) out of 15 stage III patients and 4 (27%) out of 15 stage IV patients. All samples from healthy volunteers were negative for tyrosinase. Conclusion: We confirmed high specificity and low threshold for detection of RT-PCR for tyrosinase mRNA in detection of circulating melanoma cells in melanoma patients. Circulating melanoma cells were detected in melanoma patients in all stages of the disease, but more frequently in patients with a higher stage.
