Sažetak
The main goal of this research was to isolate and identifie microorganisms growing on polychromed wooden sculpture found buried in the basement of damaged church in Gora near Petrinja. After the isolation, the microorganisms were identified by their morphological characteristic and by means of biochemical tests. Between the isolated moulds the following have been identified: Alternaria tenuis, Sordaria sp., Paecilomyces variotii and Penicillium chrysogenum. Among isolated bacteria dominated Bacillus sp. and Streptomyces sp. In order to prevent further development of the attack by microorganisms before the restoration and after it, the polychromed sculpture has to be conserved with antimicrobial substances. In this case preservative methyl-isothiazolone PBK was selected. To find optimum concentration for preservation, several mass concentrations (30-210 mg l-1) of microbiocide was prepared and tested separately on each isolated microorganism. It was found that mass concentrations of microbiocide methyl-isothiazlone PBK between 30 and 60 mg l-1 inhibited the growth almost of all isolated bacteria and moulds except the growth of mould Sordaria fimicola (180 mg l-1). So, for proper conservation of artistic object from further biodeterioration, it is suggested to apply 180 mg l-1 of PBK.
