Sažetak
Filaggrin is a predominant source of free amino acids and their derivatives largely contributing to total content of natural moisturizing factor (NMF), a complex mixture of hygroscopic compounds, which contains next to filaggrin degradation products also chloride and sodium ions, lactate, and urea. By regulating hydration and acidity in the SC, filaggrin degradation products have important function in regulation of the activity of enzymes crucial for the activity of various SC proteases involved in desquamation and lipid synthesis. Furthermore, components of the NMFs contribute to the plasticity of the skin through interaction with keratin and keratin filaments. Decreased levels of NMF are associated with a dry skin which is a major hallmark of AD. Loss- of-function mutations in the filaggrin gene (FLG) are a major determinant of NMF in the SC. In this presentation, the results of a recent study on the NMF levels in the amaXhosa population will be shown and compared with those obtained in European Caucasians.
