Sažetak
Plum pox virus (PPV) has been increasingly endangering the plum production in Croatia. Our objective was to determine the presence of PPV in autochthonous plum genotypes. We plan to subject these plum genotypes to genetic analysis in subsequent study, in order to sort out the broad array of synonyms used in plum variety designation. This would enable as to determine and describe specific genotypes (varieties) and corresponding synonyms. Ninety-three leaf and fruit samples were collected in May and August 2005, respectively, in seven Croatian counties: Osječko-baranjska, Vukovarsko- srijemska, Požesko-slavonska, Brodsko-posavska, Virovitičko-podravska, Sisačko-moslavačka and Ličko-senjska. The variety Bistrica is cultivated in all seven counties while other individual genotypes are grown in particular locations in particular counties (Bela staroverska, Bijela rana, Bjelica jajara, Brdaklija, Ceričanka, Crnica, Dabinka, Debeljara, Glodara, Kalača, Kamenjara, Komočanka, Mandalenka, Miholjčanka, Pasjara, Pintara, Plavica, Ružica, Sitna bijelica, Torgulja, Ilinjača, Trnovača and Valpovka). PPV was detected by Double Antibody Sandwich ELISA-test, using broad range polyclonal antibodies (Loewe Biochemica, Germany). Sharka virus was found in four plum genotypes ; three of them (Glodara from Dolac, Pasjara from Viljevo and Bistrica from Podvrško) contained virus both in the leaf and fruit samples, while one (Glodara from Crkvare) contained PPV only in the leaf sample. All other plum genotypes were found to be PPV-free.
