Sažetak
Pepino mosaic virus (PepMV) was first reported on greenhouse tomato (Solanum lycopersicum L.) in the Netherlands and UK. Since then this viral disease has established itself in the tomato industry worldwide and has become a significant problem. PepMV is an EPPO alert list pest. In order to verify the presence of PepMV in tomato cultivars and to enable enforcement of European Union legislation, an extensive survey among Croatian tomato growers was conducted in 2004. During a six years of survey (2004-2009), 1069 samples were tested: 1050 samples were from commercial domestic tomato greenhouse production, 17 from imported seedlings and 2 from imported fruits. Samples were analysed by enzyme-linked immunosorbent assay, DAS-ELISA and DASI-ELISA respectively. The extension with IC-RT-PCR was introduced in 2007 by the use of PepMV specific antibodies and oligonucleotide primers (UK/UK1 and TBG/UTR). The results obtained by IC-RT-PCR analysis were in complete agreement with both ELISA methods showing that one tomato fruit sample from the import was found to be infected with PepMV. These results confirmed the absence of PepMV in greenhouse tomato production in Croatia. Therefore, it becomes extremely important to use certificated seed, apply sanitary measures and conduct regular inspection monitoring in the future.
