Sažetak
Scots pine (Pinus sylvestris L.) provenances trials were established in Croatia 1982 on locality Drenovac, Forest office Duga Resa, Forest administration Karlovac as part of International Scots pine provenance experiment organized by IUFRO. The aim of the present study was to investigate the microsatellite chloroplast DNA variation in 20 European provenances and 1 autochthon Croatian provenance of Scots pine. We used seven chloroplast microsatellite loci: PCP26106, PCP71987, Pt30204, Pt71936, Pt79951, Pt45002 and Pt87268 for reasons that chloroplast genome shows lower mutation rates then nuclear genome ; it is usually uniparentally inherited and contains well studded highly polymorphic microsatellite regions suitable as tool for the study cytoplasmic diversity. The seven pairs of primers were carried out in multiplex PCR to speed up the genotyping. The results showed very large variety of haplotypes: 42 different haplotypes in 63 individuals. In almost all populations, all individuals had different haplotypes, except two individuals. A lot of unique haplotypes were detected. The total diversity was very high (hT=0, 979) as well as the average within- population diversity (hS=0, 968). Genetic differentiation was very low (GST = 0.011).
