Recent studies demonstrated that chronic rapamycin treatment extends lifespan and reduces fecundity of fruit fly Drosophila melanogaster females. The aim of this thesis was (i) to investigate the effects of increased protein concentration in diet on longevity of the fruit fly, (ii) to compare the effects of standard and chemically defined medium on lifespan and (iii) to determine whether pharmacological intervention with rapamycin late in life have beneficial effects on fly survival. In accordance with others, this study showed that increased protein component of a diet decreases lifespan. It was found that even short rapamycin treatments, of about three weeks, can extend maximum lifespan of a fruit fly to the level comparable to the chronic rapamycin treatment. This effect was observed on all diets tested, suggesting that the effect of rapamycin is at least partially independent of nutrition. Additionally, rapamycin reduced fecundity of females and releasing flies from rapamycin treatment resulted in recovery of fecundity.
