GLUCOSE REDUCES ALLOXAN INDUCED ALTERATIONS OF NOCICEPTION

According to literature data, alloxan-induced betacytotoxic effects can be prevented by prior glucose administration. We investigated the effects of peripheral (intraperiotoneal /ip/) and central (intracerebroventricular /icv/) 0.1M and 1M glucose-15 min pretreatment on centrally (icv) applied alloxan (1000 mg/kg) -induced alterations of nociception, determined by the "hot plate" and the "tail immersion" tests. Alloxan icv treatment alone induced gradual increase (Ł100% over control) of pain treshold in both tests in the course of a 3 week posttreatment period. Similar increament of pain treshold was induced by a glucose (ip/icv) treatment alone, as well. Glucose pretreatment, in general, reduced alloxan induced increase of pain treshlod in both tests. However, the effect was more consistent, pronounced with higher glucose dose (in the hot test plate even reversed to control values) and could be seen as early as 1 week following glucose icv (vs. 3 weeks following glucose ip pretreatment). It seems that glucose 15 min-pretreatment reduces alloxan icv-induced alterations of pain treshlod, with glucose icv pretreatment being more successful than glucose ip pretreatment.
