Sažetak
We studied the insulinomimetic effects of vanadium, a potent inhibitor of phosphotyrosine phosphatase on amino acid transport and on gluconeogenesis in cultured rat hepatocytes and implication of this pathway on insulin intracellular signalling to elucidate mechanisms of insulin action. In the presence of insulin and/or vanadate basal alfa-amino isobutiyric acid (AIB) uptake was increased by 100%. Basal and glucagon-stimulated gluconeogenesis was significantly decreased by insulin. Vanadate also decreased basal gluconeogenesis like insulin, but ther was no effect on glucagon-stimulated gluconeogenesis. Our data indicate that receptor tyrosine-kinaseactivity is associated withinsulin action on both AIB uptake and gluconeogenesis in hepatocytes. Morover, they suggest that a brancing of insulinsignal trnsduction occut at a post-receptor level.
