Sažetak
Research spanning the past several decades suggests that acetylcholine-containing neurons may play an important role in mediating learning and memory in animals and humans. It has been shown that a large cholinergic pathway projecting to cortical areas originates from neurons located in the ventro-medial portion of the pallidum (also known as nucleus basalis (NB) magnocellularis). Electrolytic or neurotoxic lesions of the NB in the rat bring about the degeneration of the cholinergic neurons constituting approx. 90% of its neuronal population. It was of interest to study the influence of unilateral or bilateral electrolytic lesions of the NB on the passive avoidance behaviour in rats. Anesthetized male Wistar rats weighting 200-250 g (3-4 months old) or 350-480 g (about 1 year old) were placed in a stereotaxic apparatus. The coordinates for unilateral or bilateral NB lesions for adult animals were: 1.8 mm anterior, 2.5 mm lateral and 8 mm ventral with bregma as 0. For young rats the used coordinates were: 0.7 mm posterior ; 2.7 mm lateral and 8 mm ventral with bregma as 0. The site of the lesion was histologically verified. Fifteen days later the passive avoidance task according to the procedure of Ashford and Jones (1976) was performed. A chamber with a grid floor continuously electrified was used. The central area of the floor contained a wooden platform. Each rat was placed on that platform and left in the apparatus for 3 minutes. The total time spent on the platform was recorded. A total of 4 consecutive daily sessions were given. Results of our experiments show that both unilateral and bilateral electrolytic lesions of the NB impaired passive avoidance responses in young and old rats.
