Sažetak
There is increasing evidence that non-enzymatic post-translational protein modifications might be a key event in diseases, like degenerative diseases and various protein storage diseases. Such modifications of proteins are determined by various influences which also include free radicals generated in their vicinity. In order to investigate the mechanisms, products and consequences of such modifications new methods or modifications of current ones should be made. Protein HNE-adducts ELISA is a method for detection of 4-hydroxynonenal (HNE) bounded to proteins, what is considered as the most likely form of HNE existence in living systems. HNE, a well known biomarker of oxidative stress and lipid peroxidation, has been recognized as important molecule in pathology but as well in physiology of living organisms. Since, the current ELISA has been validated for cell lysates we wanted to make it applicable also for plasma samples. Therefore, we performed a modification of current protocol and after its validation we analyzed 2 groups of samples: obese (n=62) and controls (n=15). Our results have shown higher values of HNE-Protein adducts in obese group (p<0, 0005). All samples were analysed in two separate experiments with a good correlation (r=0, 938). We also made a comparison of data obtained for HNE and malondialdehyde (MDA), another lipid peroxidation product and we found that there is a rather good correlation (r=0, 744) between these 2 parameters with significance p<0, 0005.
