Sažetak
Regardless of huge improvements in early diagnostics and therapy in cancer treatment in the last few decades, breast cancer represents leading cause of mortality and morbidity in women, mostly due to property of primary tumour to metastasize. It was revealed that metastases comprise of a heterogeneous population of cells of which the most interesting ones are stem-like cells. These cancer stem cells (CSC) possess characteristics of both stem cells and cancer cells, in that they have the properties of self-renewal, asymmetric cell division, resistance to apoptosis, independent growth, tumourigenicity and metastatic potential. CSCs are of great importance in cancer biology as they are involved in blood vessel formation, promotion of cell motility and resistance to therapies and especially to metastasis development. One of the important factors influencing the stem cell destiny is their microenvironment and their interaction with extracellular matrix (ECM). Since therapy treatments induce oxidative stress, and consequently lipid peroxidation, that cause microenvironment modifications, it is of interest to study how these modifications change CSC growth and differentiation. For that reason we wanted to investigate how will prolonged oxidative stress affect cell growth and differentiation of CSC. We have analysed the growth and differentiation of breast cancer cell line SUM 159, triple negative breast cancer cells with mammosphere forming capacity. Our results have shown that prolonged oxidative stress can affect differentiation of CSC and thus should be considered as important parameter in the research of breast cancer.
