Sažetak
Human first trimester pregnancy decidua is haevily infiltrated with bone narrow derived cells (decidual leukocytes). Besides macrophages, and T lymphocytes, large granulated lymphocytes of NK like phenotype (CD3-CD56+CD16-) are prevalent population. These cells are constitutively equipped with high levels of cytolytic protein perforin in their granules (bright+). Suspensions of peripheral blood and decidual cells, obtained by Ficoll-Hypaque gradient centrifugation, was incubated overnight in tissue culture Petri dishes to obtain non adherent (NAC) and adherent cell (AC) populations. NAC are prevalently (85-90%) nonclasical NK cells with CD16-CD56++ phenotype and large granules in cytoplasm (large granular lymphocytes-LGL). Granules of fresh isolated LGL contain high level of perforin and serine esterases. Both the content of perforin per cell, and the percent of perforin positive cells diminish after the 18h incubation in vitro, but can be prevented completely by cocultivation with syngeneic or allogeneic decidual adherent cells.
We investigated whether the peripheral blood adherent cells (PBAC) have the same effect on maintaining the perforin level in decidual and peripheral blood lymphocytes, as we observed for decidual adherent cells (DAC). Our analyses showed that the effect of 10% of DAC added are comparable with 50% of PBAC. The size and granularity of both types of cells are almost the same. PBAC appears as homogenous population of monocytes, mostly expressing typical monocyte/macrophage markers: CD14/CD45, CD11c, HLA-DR. Contrary, DAC are heterogeneous population of cells which are mostly not bone-marrow derived. Only 10-20% of them express previously mentioned monocyte/macrophage markers.
The supernatant obtained by cultivation of DAC had the same protective effect on the level of P expression as was observed by cocultivating DAC and DNAC. In our opinion these results are pointing that DAC have pivotal role in up regulation of perforin.
