A disturbance in Ca2+ homeostasis induced by hypoxia causes numerous intracellular changes including phospholipases activation and free fatty acids (FFAs) liberation. These effects start the "arachidonic acid (AA) cascade" with concomitant detrimental action on cell structure and functioning. The aim of our study was to determine the influence of indomethacin, cyclooxygenase inhibitor, on FFA pool in hypoxia-exposed rats. The study was carried out on Hannover-Wistar rats weighing 250 g. The animals of control group were intact, drug naive. All other rats were subjected to a period of oxygen deprivation hypoxia until loosing righting reflex and decapitated 15 or 60 minutes later. Two groups of animals were injected with vehicle solution. Other two groups were pretreated with 10 mg/kg of indomethacin, i. p. 30 minutes before hypoxia exposure. FFAs were quantified by gas chromatography using internal standard method. Our results demonstrate that indomethacin significantly increases total FFA level in hypoxic rats decapitated 15 minutes after the hypoxia-exposure, with no changes in free AA content. On the contrary, the increase in the brain free AA level and no change in total FFA level was observed in hypoxic animals decapitated 60 minutes after hypoxia.
