Sažetak
The osteogenesis induced by rhBMP-7 bound to inactive bone residue (IRBR) implanted in rats have been studied at ectopic and orthotopic sites. IRBR with or without BMP-7 were implanted onto calvarial defects, onto intacted calvaria and subcutaneouslly in thoracic region of rats. Implants were harvested at day 7, 12 and 21 and analysed by histology, histochemistry, histomorphometry and immunohistochemistry. The results showed differences in initial cellular response between implants from different sites. At a subcutaneous sites BMP-7-reconstructed implants induced abundant cartilage on day 7. On the contrary, BMP-7 at calvarial sites induced direct bone formation. On day 21 all implants consisted mainly of bone. In both calvarial sites bone remodeling was active and lamellar bone was already formed. Implants from partial calvarial defect completely fused with the underlying bone. The results of histomorphometry showed more pronounced and rapid osteogenessis in both calvarial implants. In conclusion we suggest that BMP-7 induced different rate of osteogenesis at ectopic vs orthotopic sites.
