Sažetak
Glycosylation, ubiquitous to both eukaryotic and prokaryotic proteins, may play an eminent role in modulation of protein function. Glycans modulate function of proteins as determinants of the membrane surface structures, interact with growth factor receptors and extracellular matrix proteins, and serve as cellular mediators for toxins and agonists. New demands for structure elucidation of complex carbohydrates arose from the functional genomics, in-gel deglycosylation and sequencing of glycoforms from complex mixtures. Here we present a new instrumental approach to sequence components of native and derivatized complex oligosaccharide mixtures from biological sources by CID-fragmentation on the new type of instrument, a MALDI Q-TOF and compare to a PSD MALDI TOF approach.
