Sažetak
For oligosaccharide analysis the acquisition of data in MS and MS/MS in the negative ion mode can provide significant advantages concerning either sensitivity or clarity of fragmentation patterns for assignment of complex structural determinants. Applied to the problem of mapping sialylated glycoconjugates, as present in complex glycosylated amino acids isolated from the urine of patients suffering from N-acetylgalactosaminidase deficiency (Schindler´s disease), a different expression of molecular species was observed in negative ion ESI and MALDI maps. In order to obtain fragmentation data for structural analysis a direct approach by (-)PSD-MALDI was addressed.
