Sažetak
Pathogenic hantaviruses in Europe cause hemorrhagic fever with renal syndrome (HFRS). HFRS is considered an immune-mediated disease with strong proinflammatory response. Hantaviruses markedly differ in the level of their pathogenicity ranging in a broad spectrum of HFRS clinical conditions. Factors influencing such differences and appropriate immune response which hantaviruses induce in humans are still poorly explored. We searched here for the main elements of the innate and adaptive immune responses in HFRS patients infected with Puumala virus (PUUV). PUUV infection generated down-regulation of anti-viral responsive gene expression in peripheral blood mononuclear cells (PBMC) in early phase of disease. Furthermore, over-expression as well as over-production of certain cytokines and chemokines has been observed. To further asses the mechanisms of Puumala virus-induced modulation of host peripheral immune response, we examined host cellular microRNA (miRNA) expression profiles of peripheral circulation in PUUV infected patients. Several miRNAs were up-regulated upon the infection. Moreover, the observed differences were also associated with the disease severity. This study suggests that disregulation of host immune response genes by the cellular miRNAs may be involved in the pathogenesis of PUUV infection.
