Sažetak
Colon cancer is the third most commonly diagnosed cancer in the world, and the second most common cause of death from cancer. Most of the colon cancer deaths result from the metastatic spread of chemotherapy resistant cells mainly to the liver. Although there is increasing evidence that a rare population of tumour initiating cells is responsible for tumour formation and maintenance, their identification in human colon cancer was first published in 2007. Colon cancer stem cells (CSC) can readily be expanded by generating colonospheres from colon cancer cell lines. Still, little is known about regulation of growth and maintenance of colon CSCs. Microenvironment was shown to be important, but often disregarded factor in modulation of CSCs tumourigenicity. Abundant with proteins susceptible to different modifications, the extracellular matrix (ECM) could hide many answers to clarify cancer initiation. As an end product of lipid peroxidation, hydroxynonenal is known modulator of important cell processes (e.g. differentiation, proliferation or apoptosis). Therefore, we have investigated the effect of hydroxynonenal modified ECM derived from normal fibroblasts on growth of colonospheres and their enrichment in CSCs.
