Beckground and Aims: Studies in rats have shown that a high intake of dietary fat induced insulin resistance. The aim of this study was to elucidate effect of troglitazone on liver insulin resistance induced by the feeding of a diet high in fat.
Materials and Methods: Hepatocytes were isolated by a modified collagense perfusion technique of Berry and Friend from normal and high-fat-fed rats. The cells were suspended to a final concentration of one million/ml M199 serum-free medium and tree ml of cells suspension were placed in 60-mm collagen coated Petry dishes. After 24 h culture medium was replaced with Hanks-hepes medium containing 2 g/L albumin and alfa-amino 14C -isobutiric acid to the final concentration of 1 mM/L, with or without 0.08 uM/L insulin, or 2 mg/L troglitazone, or 1 mM/L vanadate, or 0.1 uM/L phorbol 12-miristate 13-acetate (TPA). One hour later, after three washes with cold saline, hepatocytes were quickly frozen and an aliquot was taken for determination of protein and radioactivity.
Results: In hepatocytes obtained from rats on standard dietinsulin produced 61% increase of AIB transport. Treatment of the cells with troglitazone,produced almost same (50%) increase of basal AIB transport. This effect completely disappeared wit prolongation of treament period. On the other side troglitazone increase for almost 40% insulin-stimulated AIB transport and this enhancement of insulin action remained wit prolongaton of treatment. Almost same effect on basal transport was achived if the cells were pretreated withvanadate, but there was not any effect on insulin-stimulated AIB transport. In hepatocytes isolated from rats on high fat diet insulin effect was strongly reduced and troglitazone did not produce any increase of basal AIB transport but normelize insulin-stimulated transport.
Conclusion: These findings demostrated that troglitazone has an insulinomimetic effect on normal hepatocytes and also an insulin sensitizing effect in normal as well as in insulin resistent hepatocytes.
