Sažetak
The aim of risk assessment for chemical mutagens is to identify mechanisms of their action on DNA molecule and to put them in relation with long-term response of organism such as the development of specific cancer. It is still a common approach that the model of choice for analysing the action of some suspected mutagen is based on the primary chemical structure without sufficient utilizing the experience from toxogenetic studies. Halogenated mutagens, although often described as a more potent mutagens are difficult to distinguish biochemically from the related non-halogenated forms, that is, by analysing their formation of terminal DNA adducts and consequent mutagenic potentials. There are ever more data on non-random action of chemical and physical mutagens which originate from regional specificity of DNA molecule and regional repair intensity. In such light, well known mutagens, vinyl chloride monomer and ethylene oxide are compaired. Toxogenetics is at the stage to require stereochemistry as a potent tool for modelling new virtual space for understanding interaction between a chemical mutagen and DNA molecule.
