Sažetak
The mitotic activity of vegetative buds of Picea abies grown in vitro was compared with mitotic activity of buds grown in nature. The distribution of nuclei and the mitotic index (Mi) in the embryonic shoot was determined. Longitudinal 3 um-thin sections of fixed vegeteative buds of Picea abies from in vitro and in vivo conditions were successively cut from the beginning to the middle of the bud and examined. During winter, the cells had large, round nuclei, and no cell divisions were observed. The first cell divisions were observed with onset of spring. In two weeks, the mitotic index had increased from 3.6x10-3 to 44.1x10-3. The divisions were periclinal in the dermal layer of cells in the apex. In needle primordia, mitotic spindles were orientated in the way of primordia elongation with few anticlinal mitoses in mid-cortex. The mitotic index in the buds grown in vitro was 3.3x10-3 to 2.9x10-3. In the regions with induced adventitious buds, the divisions were anticlinal in the dermal layer of cells and anticlinal and periclinal in the layers below. In isolated adventitious buds, the mitotic index was 5.8x10-3.
