Sažetak
Introduction: Traumatic brain injury (TBI) is a major public health problem because of the high rate of mortality and long-term disability as well as due to huge cost to the victims’ families and society, both in developed and developing countries. After the initial injury caused by the direct mechanical force to the head, secondary damage develops over a period of time extending from minutes to months. There is no standard pharmacological treatment that successfully blocks the progression of the secondary injury and the current management of TBI is mainly supportive. Recently in this field, the point of interest is on multifunctional drugs that simultaneously target several detrimental factors that contribute to the secondary injury cascade included in TBI. In this presentation, overview of the current knowledge concerning the effects of multifunctional drugs in TBI will be showed. Also, the results of our research on the effects of enoxaparin, low molecular weight heparin, on different parameters of the brain damage following experimental TBI will be presented. Material and methods: Experiments were performed on adult male Wistar rats. TBI of moderate severity was induced using the lateral fluid percussion brain injury model. Enoxaparin (1 mg/kg) or vehicle were administered i.p. 1h after TBI followed by additional 7 injections administered in intervals of 6 h. Rats were sacrificed 48 h after the TBI induction. Brains were processed and analyzed by biochemical, Western blot and immunohistochemical methods. Results and conclusion: Our work suggests possible neuroprotective potential of enoxaparin in the rat TBI model.
