Sažetak
A high affinity/low capacity sodium-D-glucose cotransporter Sglt1 (Slc5a1) in the brush-border membrane (BBM) of small intestine (SI) and renal proximal tubule (PT) mediates glucose (re)absorption. A detailed localization of Sglt1 (mSglt1) along the nephron, and possibly in other mouse organs is unknown. Here we performed studies in wild type (WT) and Sglt1 knockout (KO) mice in order to detect the expression of mSglt1 protein and mRNA in various organs. mSglt1 protein was analyzed by immunocytochemistry (IC) and Western blotting (WB), whereas the expression of mSglt1 mRNA was analyzed by RT-PCR. In WT mice, mSglt1 protein was detected in the apical domain of SI, PT (where it exhibited segmental (S2>S3) differences), thick ascending limb of Henle, liver bile ducts, and pancreatic ducts. By WB, the mSglt1 protein band of 75 kDa was determined in the kidney BBM. The immunostaining and the protein band were absent in the organs of KO mice. RT-PCR data were comparable to IC data. The mSglt1 expression was not detected in the brain, spleen and skeletal muscle. This study provides new insight into mSglt1 expression in various mouse organs, thus enabling a better understanding of the role of mSglt1 in future physiological, pathophysiological, pharmacological, and toxicological studies.
