Sažetak
Embryonic stem cells are a suitable model for studying early embryonic development. The differentiation of primitive endoderm cells from the inner cell mass of blastocyst is a key event during early differentiation. Mutations in cells of the primitive extraembryonic endoderm result in defects of the embryonic development of the heart and pancreas. We monitored the development of primitive endoderm during the spontaneous differentiation of embryonic stem cells in culture. While culturing on murine fibroblasts or matrigel, embryonic stem cells begin to differentiate into primitive endoderm cells, primarly at the edges of colonies, forming the ring of Gata 4 and Gata 6 double-positive cells. Expression of pluripotency genes changed significantly during differentiation, what suggests mutually antagonistic action of these transcription factors. Pluripotency is constitutively maintained by increased expression of transcription factors OCT4. Removing exogenous FGF factors from the culture media leads to spontaneous differentiation and decreased expression of pluripotency genes. Embryonic stem cells can differentiate into primitive endoderm cells, and as such serve as a model for studying early embryogenesis events.
