Sažetak
Pseudomonas putida is an industrially interesting microorganism with an ability to degrade numerous toxic compounds. Because of it, this gram-negative rod shaped bacteria is a big potential for long term solutions of environmental problems caused by toxic compounds as a result of their use in agriculture and other businesses. It is very adaptable to different growth conditions and characterized by a big metabolic enzyme portfolio. Pseudomonas putida is also applicable for genetic manipulation since its genome has been sequenced and it is metabolically very versatile. Pseudomonas putida strain VLB120∆C is a solvent tolerant strain and as such could have numerous applications in two phase biotransformations. Pseudomonas putida strains KT2440 and VLB120∆C were cultivated in different reactors. Batch, fed- batch and continuous experiments were investigated in shake flasks and laboratory bioreactor. During batch cultivation, the influence of two different growth media, M9 and M12, for microorganism cultivation were investigated by experiments with both strains. Best results in shake flasks were achieved in experiments with starting concentration of glucose γ = 15 gdm3 by cultivating bacteria on M9 media for both strains. For strain VLB120∆C highest specific growth rate of  = 0.837 h-1 was recorded with final maximum biomass concentration of cX = 11.58 g dm-3 when cultivated in a batch reactor with DO concentration of 30% and pH 7. Same media was used in fed- batch experiments where highest growth rate of  = 0.599 h-1 and maximum biomass concentration of cX =21, 85 g dm-3 were achieved in experiment with specific glucose feed rate of 9.5 g h-1 dm-3. In experiments performed in continuous reactor for different retention times, M12 growth media with 15 g dm-3 glucose was used as inlet stream. The experiment was carried out for 5 days and biomass concentration of cX = 7.50 g dm-3 was achieved for retention time of  = 20 h. There was no significant difference between the two investigated strains when cultivated on glucose.
