Sažetak
Efficient microbial conversion of carbohydrates from renewable feedstocks makes a sound basis for development of sustainable bioprocesses for biochemicals production. Utilization of the renewables shifts typically used submerged bioprocesses towards (semi-)solid state bioprocesses and it requires optimization and implementation of specific types of biocatalyst, bioreactor and analytical framework. Production of D-/L-lactic acid from several carbohydrates by three selected lactic acid bacteria strains was characterized in submerged and semi-solid state model systems. Different suspensions of starch (S0 = 100 g L- 1) and starch-containing feedstock (S0 = 60 and 100 g L-1) were used as media within the semi- solid state model systems in which the D-/L- lactic acid was produced through simultaneous saccharification and fermentation by the lactic acid bacterium strain that possess amylolytic and fermentative activity. Functioning of two different types of lab-scale bioreactors within the semi-solid state model systems was tested. Gravimetric, colorimetric and chromatographic analyses were adjusted in the analytical framework that was built for monitoring of the submerged and the semi-solid state bioprocess. High efficiency of the bioprocess for the D-/L- lactic acid production by the three strains was achieved. Availability of certain growth factors in starch-containing feedstock suspension in demineralized water limited amylolytic and fermentative activity of the bacterial strain and the efficiency of the semi-solid state bioprocess thereof.
