Sažetak
"Mal secco" is a pathology of Citrus plants caused by mitosporic ascomycota Phoma tracheiphila. This pathogen produces o phytotoxic complex called "malseccin", a 93KDa glycoprotein released in O. tracheiphila culture filtrates and in the infected host plant. The effects of the malseccin complex on Citrus leaves can be evidenced only under light conditions and this factor seems to play a pivotalrole in the disease development. Physiologicalexperiments have evidenced that the presence of filtrates of P. tracheiphila grown in presence or absence of lyophilised twigs and leaves (LTL) of tolerant (monachello) or susceptible (femminiello) lemon varieties trigger the activity of superoxide dismutase, catalase, glutathione peroxidase involved in oxidative burst defence.The presence in in the medium of tollerant Citrus LTL induces in the partogen oxidative stress which affects its ability to produce 60KDa proteins and polygalacturonase, laccase, Mn dependant peroxidase. On the contrary if P. tracheiphila was grown in the presence of susceptible Citrus LTL an high stimulation of hydrolytic activities occured. Considering that 6 peptides sequenced by Fogliano et al. (1998) from Pt60 glycoprotein cleaved by tripsin treatment several couples of degenerated primers were created. The conceptual translation of several fragments (40 amplicons) obtained showed significant homologies (>50-60%)with irom membrane transporter, NADPH-dependent reductase and monoammine oxidase, enzyme that catalyses the oxidation of ammine and releases as by-products NH4+ and H2O2. A subsequesnt MALDI - TOF/TOF analysis on extracellular proteins of P. tracheiphila was carried out in order toevaluate the presence of Pt60-like proteins in the SDS-PAGE separated bands.This analysis has shown that a complex of proteins with different functions, among which monoammine oxidase, is present in the single SDS-PAGE 60KDa band. Rt-PCR analysis carried out on P. tracheiphila grown in presence or abscence of susceptibil and tollerant LTL indicated that the fragment coding for monoammine oxidase is expressed in different conditions. Among severeal factors involved in "mal secco"monoammine oxidase could play a pivotal role together with hydrilytic enzymes in the development of the disease. The physiological and molecular results higly underline the role of oxidative stress in the Phoma tracheiphila-Citrus limon interaction.
