Sažetak
The ACS:180 plus system is an automated chemiluminescence system that measures the amount of light emitted during a chemiluminescent reaction involving the ACS assays, using acridinium ester as the label and paramagnetic particles for the solid phase. Hormones (T3, T4, FT3, FT4, TSH, LH, FSH, prolactin, total HCG, estradiol, progesterone, testosterone, cortisol), drugs (digoxin, vancomycin, phenobarbital, theophylline, phenytoin, gentamycin), tumor markers (CEA, AFP, ferritin, PSA), IgE and vitamin B12 were evaluated according to the guidelines of ECCLS and of SFBC. The reagents and test definitions were supplied by Ciba Corning. Analytical evaluation was done by measuring intra- and inter-assay precision, accuracy, carry-over, linearity and comparison. Comparison study was made with RIA, IRMA and DELFIA methods. Intra-assay CV for hormones, drugs, tumor markers, IgE and vitamin B12 was 0.42%-3.56%, 0.38%-2.87%, 1.34%-4.81%, 2.45%-3.17% and 2.56%-4.00%, respectively. Inter-assay CV (within 10 days) for hormones, drugs, tumor markers, IgE and vitamin B12 was 1.99%-5.66%, 2.34%-5.45%, 3.43%-7.21%, 4.12%-5.77% and 4.11%-4.99% respectively. Sample to sample carry-over was negligible(K less than 0.01%). Study on dilutions showed good linearity for the analytical range. Results of the comparison study showed satisfactory correlation coefficients for hormones r=0.840 to 0.996, for drugs r=0.993 to 0.998, for tumor markers r=0.961 to 1.000, and for IgE r=0.978. The evaluation showed the ACS:180 PLUS to be a precise, accurate and reliable instrument in laboratory routine.
