Sažetak
A novel gene encoding protein with MATH-BTB domains was identified from cell-type specific cDNA libraries of wheat zygote and early proembryo. This gene, named TaMAB2, was transiently expressed in maize suspension cells and was found to localize around the nucleus of interphase cells. Truncating the MATH and BTB domains has a differential effect on subcellular localization suggesting that MATH domain is responsible for specific localization of TaMAB2 protein. The GFP-tagged wheat protein was also followed through egg cell/early embryo stages in Arabidopsis. Yeast-two-hybrid experiment showed that the protein interacts with itself and cullin 3 what suggests that it acts as part of ubiquitin E3 ligase complex. The protein appears to have two interesting properties: it is asymmetrically inherited and it appears to mark the plane of zygote cell division by degradation of substrate proteins.
