Sažetak
Variation of the most common type found in cervical cancer, the Human papillomavirus type 16 (HPV16), has been extensively investigated in all viral genes except the E1. We have recently thoroughly described a specific HPV16 variant containing 63-nucleotide duplication at the position 1374 within the E1 gene (E1-1374^63nt) that is relatively common being found in about 10% of HPV16 positive cervical samples in the Croatian population [I. Sabol et al. PLoS One 2012]. In addition, the HPV16 E1-1374^63nt variant was significantly more associated with low-grade cervical lesions compared to the reference HPV16 (E–r ; European prototype) [I. Sabol et al. J Med Virol 2008]. Therefore, in this study, we assessed the viral genome integration into host cell genome of the HPV16 E1-1374^63nt variant using two methods ; the DIPS (Detection of Integrated Papillomavirus Sequences) [M. Matovina et al. Gynecol Oncol 2009], and the rli-PCR (reverse ligation inverted PCR) [M. Kalantari et al. Diagn Mol Pathol 2001]. The integration analysis was done on 77 cervical DNA samples. In the majority of cases (50/56 ; 89.3%), only episomal viral genomes were found and there were no sample with integrated viral genome alone. However, in 6 of 56 (10.7%) amplicons of unexpected size indicate potentially integrated DNA. There were 15.4% and 11.5% amplicons of unexpected size in LSIL and HSIL samples, respectively, but no statistical differences were found. Thus, it appears that E1-1374^63nt duplication and variants that contain this duplication do not have a significant effect on HPV integration frequency in HSIL and LSIL samples.
