Sažetak
Proteinaceus elicitor secreted by phytophatogenic oomycete Phythophthora cryptogea, β-cryptogein, induces hypersensitive response and systemic acquired resistance in plants against some pathogens. In order to mimic pathogen attack that could lead to generalized stress response, and thus improved antioxidant status of the roots, Coleus blumei hairy roots harboring crypt gene were generated. Agrobacterium rhizogenes was employed to insert synthetic crypt gene, encoding cryptogein, under the control of alcohol-inducible promoter. The expression of β-cryptogein in C. blumei hairy roots was controlled by application of 0.1% and 1% ethanol. Antioxidative status was determined by NADPHoxidase, catalase and peroxidase activities and by measuring lipid peroxidation products and H2O2 contents. Transgenic roots for analysis were harvested on the 3rd, 7th and 14th day after ethanol treatment. After two weeks, ethanol-induced expression of β-cryptogein caused significant increase in catalase, peroxidase and NADPH-oxidase activities and increase in content of lipid peroxidation products. Increased enzyme activities coincided with decreased H2O2 content. These results indicated that the inducible expression of the β-cryptogein in transgenic roots, positively correlated with the activities of catalase, peroxidase and NADPH-oxidase, could improve plant defense response to biotic and abiotic stress.
