Sažetak
Due to their high consumption, pharmaceuticals along with their metabolites are being continuously introduced into the environment, mainly through excreta, disposal of unused drugs or directly from pharmaceutical discharges. They are present in low or very low concentrations ; this, together with their different physico-chemical properties and the complexity of environmental matrices, makes their determination difficult. It is therefore necessary to develop sensitive, selective and easily reproducible methods for their determination in different environmental samples. Solid phase extraction (SPE) is currently the most widely used enrichment technique for pharmaceuticals residues in water samples. The optimization of an SPE procedures implies the selection of the most appropriate sorbent, the determination of the volume of the sample to load and determination of the nature and volumes of solvents first used to wash the column and then to elute the analyte. The choice of sorbent is the key point in SPE because it can control parameters such as selectivity, affinity and capacity. The main goal of this paper was to obtain the best extraction efficiency for investigated pharmaceuticals. The pharmaceuticals studied were fluoroquinolones (enrofloxacine, norfloxacine and ciprofloxacine), sulfonamides (sulfadiazine, sulfamethazine and sulfamethoxazole) and their metabolites (N-acetylsulfadiazine, N-acetylsulfamethazine and N-acetylsulfamethoxazole). Different SPE materials (Strata X, Strata C18, Agilent C8, Oasis HLB) were used for obtaining the best cartridges for all pharmaceuticals investigated. Extraction efficiency was determined by HPLC-MS-MS. Separation and quantification of the selected pharmaceuticals were carried out under gradient elution of mobile phase (0.1% formic acid in water and 0.1% formic acid in acetonitrile) based on Synergy Fusion C18 embedded column (150 mm×2.0 mm, particle size 4 µm (Phenomenex)).
