Sažetak
In vitro culture of single node microcuttings is often employed for producing large amounts of homogeneous plant material for cryopreservation experiments. In this work, we investigated the effect of culture duration of grapevine microcuttings on medium with zeatine riboside (ZR) on shoot tip recovery after cryopreservation. Single node microcuttings of grapevine cultivar Portan were placed on ½ MS medium + 1 µM ZR with transfers to new medium every 4 weeks, and shoot tips were excised after 4, 8, 12 and 16 weeks of culture. Control explants consisted of shoot tips sampled from microcuttings cultured for two months on ½ MS devoid of ZR. The droplet-vitrification technique was employed for cryopreservation of shoot tips. After excision, shoot tips were placed for 24 h on medium containing 0.3 M sucrose, treated for 20 min with loading solution containing 1.2 M glycerol + 0.4 M sucrose, dehydrated with the PVS2 vitrification solution for 25, 50 or 75 min, cooled rapidly in PVS2 droplets placed on aluminium foils, rewarmed rapidly by immersion of foils in unloading solution with 1.2 M sucrose for 20 min, then transferred on standard medium for recovery. After cryopreservation, 30-33 % recovery was obtained for shoot tips sampled from microcuttings cultured for 0, 4 and 8 weeks on medium with ZR. No recovery was noted with shoot tips sampled from microcuttings cultured for longer periods on ZR medium. These results underline the importance of selecting explants in an optimal physiological state for cryopreservation experiments.
