Sažetak
Bone morphogenetic proteins (BMPs)/osteogenic proteins (OPs) are members of the TGFb superfamily and act in a wide range of tissues in mouse development. BMPs are expressed and coordinated in activity in every cell population during kidney development. Several BMPs are expressed in embryonic kidneys and fetuses lacking BMP-7 gene die of kidney failure shortly after birth. BMP-7 induces E11 metanephric mesenchyme in vitro and initiates expression of specific nephrogenic markers. In this study, we characterized the temporal expression of BMPs -2-6 and OP-1 (BMP-7) from E11 of mouse embryonic development up to six months of postnatal life by RT-PCR and by in situ hybridization. Results showed that all tested members of BMP family were expressed in kidney at early stages of development. BMP-2, BMP-6 and OP-1 were expressed in E11 of mouse development, with OP-1 being the most abundant. Subsequently, BMP 2-4 and OP-1 were downregulated, while expression of BMP-6 increased in time. The colocalization of BMPs suggests that members of this family can substitute functionally for each other. In postnatal life, expression of BMP-2 was strongly reduced, while BMP-4, BMP-6 and OP-1 were expressed throughout the period of 1.5 years supporting their role in maintaining kidney structure and function after birth. We suggest that BMPs might recapitulate events of embryonic development during kidney injury.
