Sažetak
Involvement of endogenous abscisic acid (ABA) and indole-3-acetic acid (IAA) in stress-induced somatic embryogenesis of pumpkin (Cucurbita pepo L) was analyzed using two different stress treatments: (a) medium with 1 mM 2, 4-D, and (b) hormone-free medium with 1 mM NH4+ as the sole source of nitrogen. Both treatments promote proliferation of proembryogenic tissue masses, while further embryo development can be obtained only after transfer to regeneration medium (hormone-free medium with optimal supply of nitrogen). Endogenous hormone contents were determined after 7 and 21 days of treatment, and 3 and 5 days after the transfer on regeneration medium. Considerably higher endogenous ABA level was observed in embryogenic tissue cultured in presence of 2, 4-D when compared to NH4+-medium (8.3 or 1.5 pmol/g fresh weight). Seven days after the transfer on regeneration medium ABA content additionally increased, regardless to the previously applied stress treatments. On the other hand, content of endogenous free IAA and IAA-Asp was significantly higher in presence of NH4+- then 2, 4-D (417 or 77 pmol/g fresh weight). After the transfer from NH4+ or 2, 4-D-medium to the regeneration medium, level of endogenous IAA declined. Results suggest that 1 mM NH4+ as the sole source of nitrogen increases level of endogenous IAA, required for the acquisition of embryogenic capacity in pumpkin. Decrease in endogenous IAA content was connected with the later stage of somatic embryo development.
