Objectives: Kobuviruses comprise a group of small, non-enveloped, positive-sense, single-stranded RNA viruses that have a zoonotic potential. So far they have been isolated from humans, cattle, sheep, goats, swine, dogs and probably bats. Bovine kobuvirus belongs to the Picornaviridae family, genus Kobuvirus along with Aichi virus and several candidate species. Worldwide geographical distribution is quite possible ; however, available data describes the presence of Bovine kobuvirus in Japan, South Korea, Thailand, Hungary, Belgium and The Netherlands. Generally, kobuviruses cause an inapparent course of infection, though clinically observable enteric disease is also described. Recent research studies indicate the possibility of detecting the viral RNA of Bovine kobuvirus in stool samples of diarrhoeic as well as healthy cattle. However, the epidemiological relatedness to enteric disease has only been confirmed for Aichi virus infections in humans, whereas kobuviruses isolated from animals still remain undetermined as actual causative agents of enteric disease. In this study we demonstrate the first detection, genotyping and phylogenetic analysis of Bovine kobuvirus strain isolated from diarrhoeic cattle in Croatia. Methods: In March 2011 our Laboratory received two samples of the small intestines originating from calves from a large industrial farm, to exclude bovine viral diarrhoea (BVD). In addition, we collected six more stool samples in October 2011 from healthy cattle of different age, originating from small and medium sized farms. All farms are located in the Eastern part of Croatia. On these samples we performed RT-PCR employing previously published generic primers to detect the partial 3D region of Bovine kobuvirus genome. PCR products were sequenced directly in both directions and phylogenetic analysis was conducted by using MEGA 5 software. Results: Two out of eight samples, tested positive for Kobuvirus by RT-PCR showing a band of expected size. Both positive samples originated from dead diarrhoeic calves bred on the large industrial farm and were negative for BVD. Phylogenetic analysis confirmed the presence of Bovine kobuvirus. Croatian isolate BoKoV/T1/Cro, clustered together with other previously reported Bovine kobuviruses. Conclusion: Bovine kobuvirus is present within the cattle population in Croatia which supports the statement of its probable wider distribution. Our results indicate its possible relationship with enteric disease, as it was detected only in diarrhoeic calves. Phylogenetic analysis of Croatian strain revealed that it is genetically closely related to other known Bovine kobuviruses. More comprehensive research is needed to get a better insight in Bovine kobuvirus prevalence among healthy cattle in Croatia and to resolve its importance as the causative agent of enteric disease.
