Sažetak
Because of its potential for degradation of organic pollutants, there are efforts to grow bacteria Pseudomonas putida in large amounts, in order to be used in bioremediation processes of cleaning soils contaminated with toxic compounds commonly used for agricultural purposes. In this work two strains of bacteria Pseudomonas putida, KT2440 and VLB120∆C, have been cultivated on two different nutrient media, M9 and M12, in experiments performed in the shaking flasks and in the batch bioreactor with aeration. The main goal of the investigation was to determine the affinity of growth of different strains on certain nutrient medium, . Additionaly influence of pH and dissolved oxygen concentration on biomass growth and glucose consumption was analysed. The results show that the largest biomass growth in flasks was observed in experiment performed on M9 medium, while the largest increase of biomass was achieved in experiments performed in the bioreactor on M12 medium. It was also showed that for experiments performed in flasks, biomass growth is inhibited due to low pH-value and probably due to the lack of dissolved oxygen. In flasks experiments, the highest yield of biomass was achieved on M9 medium for strain KT2440 and it was YX/S = 0.1912 g g-1. A mathematical model of the processes was developed, and parameters of Monod kinetic model were estimated (KS = 1.0360 ± 8.5213 g dm-3 and µMAX = 0.4609 ± 0.2752 h-1). In the experiments performed in batch bioreactor with aeration highest biomass yield of YX/S = 0.7450 g g-1 was obtained on M12 medium for KT2440 strain. Also for this experiment, a mathematical model of the process was developed and parameters of Monod kinetic model were estimated (KS = 9.0442 ± 16.5264 g dm-3 and μMAX = 0.7245 ± 0.5625 h-1).
