Sažetak
Oilseed rape (Brassica napus ssp. oleifera) is increasingly important cruciferous crop used for consumable oil and biodiesel production. Plants showing virescence, phyllody, witches’ broom, small and seedless siliques symptoms along with some stem necrosis, leaf chlorotic spots and necrotic changes along the veins were collected from the field and investigated for the presence of phytoplasmas and viruses. Total nucleic acids were extracted from leaf midribs, stem phloem or inflorescence tissue for PCR-RFLP identification of phytoplasma 16S rRNA gene. Generic phytoplasma primer pairs P1/P7 and R16F2n/R2 were used for direct and nested PCR, respectively. Amplification products from nested PCR were singly digested with restriction endonucleases Tru1I (MseI) and TasI (Tsp509I) allowing the identification of phytoplasma Aster Yellows (‘Candidatus Phytoplasma asteris’) ribosomal subgroup I-B. The amplicon of approximately 1800 bp from the direct PCR was cloned and sequenced. Phylogenetic analyses of 16S rDNA corroborated RFLP results showing that the isolated strain was very closely related to other strains assigned to the 16SrI-B subgroup. Classification of identified phytoplasma was additionally confirmed by in silico restriction analyses using iPhyClassifier on-line tool. Biological testing was performed by mechanical inoculation of sap from symptomatic plants on a wide range of experimental plant host species. The host range and biological characteristics suggested the infection of oilseed rape by Turnip mosaic Potyvirus (TuMV). Along with the virus amplification in propagation hosts B. rapa and Nicotiana benthamiana, the serological TuMV identification was performed by using polyclonal antibodies, as well as PCR experiments on total plant RNA employing specific TuMV-coat protein primers. The viral amplicon sequencing and phylogenetic analysis confirmed the TuMV identification and revealed its affiliation to the worldwide distributed B-group. This is the first report of mixed AY phytoplasma and TuMV infection in Brassica sp. Nevertheless, having in mind the worldwide distribution and pathogenicity of these two pathogens, both the occurrence of this mixed infection and its negative impact on the plant production are surely underestimated.
