Sažetak
Tobacco industries produce large quantities of solid and liquid wastes. These wastes pose an important environmental problem, as some main components are harmful and toxic. The aim of this work was to isolate and identify the nicotine degrading microorganisms during composting of tobacco waste. The bioremediation process for detoxification of waste was carried out in a column reactor at an airflow rate of 0.4 L min-1 kgVS-1. During 19 days were monitored the concentrations of nicotine and number of CFU in the samples taken from reactor. After 19 days 89.8% of nicotine conversion was obtained. A nicotine-degrading bacterium, strain FN, was isolated from composting mass and identified as Pseudomonas aeruginosa based on morphology, tests by API 20 NE set, 16S rDNA sequence and phylogenetic characteristics. To verify that isolated Pseudomonas aeruginosa FN is the true nicotine degrader, batch experiments using tobacco leachate were carried out. It was confirmed that strain FN possess considerable ability to degrade nicotine with simultaneous COD removal. Monod kinetics model for single substrate was solved to obtain substrate degradation rate and half saturation constant.
