Sažetak
Electroanalytical methods, in particular voltammetric techniques, are well established in pharmaceutical and biomedical analysis. Cyclic voltammetry is very suitable to investigate the electrochemical properties of a drug to give insights to metabolic fate or in vivo redox processes and pharmacological activity. Square wave voltammetry has been particularly useful for trace measurements of electroactive compounds in body fluids and tissues as well as quality control of pharmaceutical dosage forms without the necessity for sample pretreatment. Parkinson’s disease is a progressive, neurodegenerative disorder primarily affecting dopaminergic neuronal systems, with impaired motor function as a consequence. Ropinirole is a non-ergoline dopamine agonist used in the treatment of Parkinson’s disease. In order to characterize the electrochemical oxidation behavior of ropinirole, cyclic and square wave voltammetry were carried out in various supporting electrolytes. The voltammograms recorded in acidic media exhibited a single well-defined oxidation peak at 1.28 V. Drug molecule was irreversibly oxidized resulting in the formation of a couple with a reduction and re-oxidation peak at much lower potential. The effect of pH on the peak potential and intensity were investigated. The shift of the peak potential to less positive values with increase in pH indicated the involvement of proton in the electrode reaction before the electron transfer rate-determining step. The plot of the peak potential vs. pH showed a straight line between pH 2 and 11 (Ep(V) = 1.51−0.064 pH). The mechanism of the oxidation process will be discussed. Scan rate studies showed that the process was diffusion controlled. The variation of logarithm of peak current as a function of logarithm of scan rate in the range of 10–500 mV/s yielded a straight line with a slope of 0.58, very close to the theoretical value of 0.5, which is expected for an ideal reaction of solution species. Square wave voltammetric technique was developed for the determination of ropinirole in bulk form. In 0.1 M H2SO4, the linearity of the calibration curve was obtained in the range of 2×10−5 to 5×10−7M using glassy carbon electrode.
