Sažetak
Trophoblast cells in the closest contact with decidualised endometrium secrete progesterone in a high concentration. Progesterone and its mediator Progesterone Induced Blocking Factor (PIBF) support Th2 type cytokines production in decidua suppressing cell-mediated immunity of decidual lymphocytes (DL). DL are predominantly non-classical NK cells of CD16-CD56 bright phenotype abundantly endowed with cytolytic mediator-perforin. Progesterone and PIBF containing supernatant in vitro indirectly block upregulation of perforin expression in DL cocultured by decidual macrophages and decrease cytolytic activity of DL. Cytotoxic activity of progesterone stimulated DL was tested against K562 cell line in 2 hr cytotoxicyty PKH 26 (red) test. Th1 cytokine IL-15, wich shares many biological properties with IL-2 is also abundantly present in first trimester normal human pregnancy decidua. IL-15 in vitro strongly upregulates perforin expression in DL, while does not affect their cytolytic activity against NK target K562 cells. As DL are in vivo under both progesterone and IL-15 influences we analysed in vitro the interference of progesterone/PIBF and IL-15 stimulation to perforin expression. Purified PIBF in concentrations 1, 2 and 4 g/ml were used. The experiments which are in progress are showing that a complex cytokine and hormone network operates at the interface.
