Sažetak
It has been suggested that decidual NK cells (dNK) besides functions in the defence against virally infected and transformed cells may contribute also to the control of cytotrophoblast invasion of the placental bed during early pregnancy. For these functions dNK cells are abundantly endowed with the perforin (Pbright), cytolytic mediator at the molecular level. Pregnancy has been defined as a Th2 phenomenon, with a prevalence of Th2 type cytokines at the maternal-fetal interface. Since cytolytic activity is regulated primarily by Th1 type cytokines, and suppressed by Th2 type cytokines, the question is which cytokines at the interface have a dominant role. IL-2 has not been detected in the normal implantation site, but IL-15 which shares many biological properties with IL-2 has been abundantly expressed in the placenta. IL-12 is expressed in placenta in small quantities and it is a cytokine capable of inducing differentiation of naive T cells to Th1 phenotype. The aim of this study was to investigate the role of Th1 cytokines (IL-2, IL-12, IL-15) in the control of NK cytotoxicity of decidual lymphocytes (DL). Cytolytic activity of unstimulated and Th1 cytokine stimulated dNK cells was tested against K562 cell line in a 2 hr cytotoxicity PKH 26 (red) test. The frequency of P+ cells and P content per cell were measured simultaneously. Cytolytic potential of DL is highly increased by IL-2 (1000 i.u./ml) and IL-12, but surprisingly suppressed by IL-15. This suppression is released in the combination of other Th1 type cytokines. These results are presenting that P mediated cytotoxicity under the influence of Th1 type cytokines might have an important role in the protection of the feto-placental unit against both maternally acquired and local infections at the interface.
